Research Achievement | UNILIPO's study on "Ionic liquid microcapsules for the topical delivery of pterostilbene" has been successfully published in a journal ranked in the SCI Zone 1

Why develop?

Pterostilbene liposomal microcapsule technology

  • vantage

■ Anti-aging

Pterostilbene, the key active ingredient in [PTEROCARPUS MARSUPIUM BARK EXTRACT], is a natural dimethylated structural analog of [RESVERATROL] and has been clinically proven to have strong anti-aging properties.

■ Whitening and Spot Reduction

It also has a dual "de-pigmentation" effect by inhibiting tyrosinase and breaking down melanin, making it even more effective in whitening and spot reduction compared to compounds such as 377. For example, in the tyrosinase activity inhibition test, [Pterostilbene] reached 91 times the efficacy of [KOJIC ACID]!

  • drawbacks

However, through our practical application in Pterostilbene Essence 2.0, we also found that pterostilbene has some unavoidable issues.

■ Decomposition under light exposure

In previous stability tests at Research & Knowledge Laboratory, higher concentrations of pterostilbene were found to degrade under conditions of high temperature or light exposure, and the color may darken, affecting the user experience.

■ Weak transdermal ability

Laboratory test results showed that without encapsulation, the skin retention of free pterostilbene within 8 hours remained at 5-6 µg/cm². Although this reaches an effective dosage, it requires increasing the amount added or using other permeation enhancers to promote absorption, which may increase skin irritation.

After research and discussion, UNILIPO has encapsulated and treated pterostilbene with ionic liquid to improve its stability and water solubility.

Comparative experiments show that pterostilbene treated with encapsulation and ionic liquid demonstrates significant improvements in in vitro stability, cytotoxicity, transdermal absorption rate, antioxidant, anti-wrinkle, and whitening effects compared to free pterostilbene.

Therefore, we call this new ingredient [Pterostilbene Liposomal Microcapsules]

Nano liposomal encapsulation of pterostilbene can achieve high stability and is non-cytotoxic.

Experimental evaluation shows that Mat-IL-MicCap@PTB (pterostilbene liposomal microcapsules) maintained 95% of the PTB content even after 90 days at high temperature (45°C) and low temperature (-25°C).

This unequivocally proves that Mat-IL-MicCap@PTB (pterostilbene liposomal microcapsules) exhibits high stability!

DLS measurement and zeta potential indicate that the resulting microcapsules have a uniform particle size distribution without aggregation into large particles. A higher zeta potential suggests greater stability.

✅ Therefore, pterostilbene liposomal microcapsules exhibit good stability in aqueous solution.

The laboratory used human kidney fibroblast (HKF) and G361 cell lines as models to evaluate the cytotoxicity of Free PTB (free pterostilbene) and Mat-IL-MicCap@PTB (pterostilbene liposomal microcapsules)

✅ It is proven that Mat-IL-MicCap@PTB (pterostilbene liposomal microcapsules) has very low toxicity to HKF cells, with almost no harmful effects on healthy cells.

✅ Mat-IL-MicCap@PTB (pterostilbene liposomal microcapsules) has a significantly greater inhibitory effect on G361 cells than Free PTB (free pterostilbene), increasing contact between PTB and cells and enhancing the water solubility of pterostilbene.

Significant enhancement in transdermal delivery permeability and long-term maintenance of high concentration.

Comparison of the efficiency and cumulative permeability of Free PTB (free pterostilbene) and Mat-IL-MicCap@PTB (pterostilbene liposomal microcapsules) penetrating the skin after 4 and 8 hours of incubation.

❌ Free PTB (free pterostilbene) struggled to penetrate the skin after 4 hours of incubation.

✅ After 30 minutes of application, Mat-IL-MicCap@PTB (pterostilbene liposomal microcapsules) exceeded more than twice the amount of free pterostilbene; the microcapsules can improve the penetration efficiency of pterostilbene. The skin retention at different time points was all higher than that of free pterostilbene.

✅ After 8 hours of reaction, the skin retention rate of Mat-IL-MicCap@PTB (pterostilbene liposomal microcapsules) was 7.7 times higher than that of free pterostilbene.

Free radical elimination

Enhanced antioxidant capacity

At the same concentration, compared to free pterostilbene

✅ Pterostilbene encapsulated in liposomes significantly increased its ability to eliminate oxygen free radicals (including DPPH and ABTS) by 2.5 times and 33 times respectively (calculated as a 90% clearance rate).

Significant improvement in whitening ability

At the same concentration, the encapsulated pterostilbene increased melanin inhibition rates by 4.8% and 11.9%, respectively, resulting in better whitening effects.

Anti-wrinkle and firming

Promotes the synthesis of type I collagen

At concentrations of 0.0156%, 0.0313%, and 0.0625% (v/v), pterostilbene encapsulated in liposomes significantly increased the amount of type I collagen in fibroblasts compared to free pterostilbene.

✅ This indicates that pterostilbene encapsulated in liposomes possesses better anti-wrinkle and firming effects.

After using pterostilbene encapsulated in liposomes for 14 days, wrinkles were significantly reduced by 8.27%, 12.95%, and 48.07%; after 28 days, wrinkles were significantly reduced by 14.60%, 18.01%, and 55.78% (figures a, b, c).

✅ The area, quantity, and length of crow's feet showed improvements.

✅ Skin firmness along the jawline improved after 14 and 28 days (figure d).

The VISIA skin analysis system showed that after using pterostilbene encapsulated in liposomes for 28 days:

✅ There was a significant reduction in the number and area of crow's feet, inhibition of melanin production, brighter skin tone, and improvement in dark spots.

Pterostilbene encapsulated in liposomes increases the firmness and elasticity of the skin, inhibits the formation of wrinkles, and does not produce adverse effects. It is gentler and more suitable for the skin of people in the country.

✅ The final research demonstrates that pterostilbene encapsulated in liposomes has high efficacy in antioxidant, anti-wrinkle, whitening, and skin repair functions!

Currently, the UNILIPO team has applied [Pterostilbene Liposomal Transdermal Microcapsules] in products such as VC/VA/Pterostilbene essence, and it will also be used in many upcoming new products.

The UNILIPO team has always been at the forefront of technology, consistently adhering to extensive research experiments and data support, specializing in the study of truly effective core ingredients and formulation techniques to rejuvenate cells at the cellular level.

In the future, we will share more research findings with you. Please stay tuned.
